Hello, I have had the same issue. I've got the solution, so I've picked up the first exotic mission with Arthur back in chapter 4. After that I picked up all plants and Orchids with John (and got 15 Lady Slipper Orchids) for the exotic mission buut I found out it is not counted towards the challenge until you completing the first exotic mission. Also, check out the LSO at the entrance of the cave northwest of Wallace Station if you haven't already. From my experience, orchids respawn after a few in-game days. 3 sounds about right, maybe wait another day.
